چکیده

Many studies in vitro have implicated cytochrome oxidase and monoamine oxidase in the metabolism of epinephrine and other cateehol amines (2). However, no evidence for the participation of cytochrome oxidase in the metabolism of epinephrine in viva has been obtained. Furthermore, several investigations have indicated that monoamine oxidase plays only a minor role in the inactivation of epinephrine (3-5). Recently, Armstrong and McMillan have shown that a major metabolic product of norepinephrine in man is 3-methoxy-4-hydroxymandelic acid (6). Work in this laboratory has also demonstrated that O-methylation of epinephrine and other catechols precedes deamination in viva (7, 8). These observations suggested the presence of an enzyme that can carry out the 0-methylation of catechol amines. This paper describes the properties of an enzyme that transfers the methyl group of S-adenosylmethionine to the hydroxyl group in position 3 of epinephrine and other catechols.

Dynamic aspects of enzymatic O-methylation and -demethylation of catechols in vitro and in vivo.

The importance of 0-methylation of catecholamines and other catechols as a metabolic pathway has been amply shown (l-4). The isolation of “Substanz Y,” a p-O-methylcatechol derivative from beef adrenal glands (5), and the occurrence of p-o-methylcatechol derivatives in urine (6) are suggestive of p-o-methylation in viva. Previous studies (7) have shown that in vitro the enzyme 0-methyltransfera...

Inhibition of DNA methylation by caffeic acid and chlorogenic acid, two common catechol-containing coffee polyphenols.

We studied the modulating effects of caffeic acid and chlorogenic acid (two common coffee polyphenols) on the in vitro methylation of synthetic DNA substrates and also on the methylation status of the promoter region of a representative gene in two human cancer cells lines. Under conditions that were suitable for the in vitro enzymatic methylation of DNA and dietary catechols, we found that the...

Methylation of halogenated phenols and thiophenols by cell extracts of gram-positive and gram-negative bacteria.

O-methylation of 2,6-dibromophenol was studied in cell extracts prepared from Rhodococcus sp. strain 1395. O-methylation activity was enhanced by the addition of S-adenosyl-l-methionine but was not affected by the addition of 5-methyltetrahydrofolate nor by up to 10 mM MgCl(2) or EDTA. By using 2,6-dibromophenol, 4,5,6-trichloroguaiacol, and pentachlorothiophenol as the substrates, O-methylatio...
